Abstract

Soluble proteins leached from pieces of natural rubber latex gloves immersed in water were purified by acid precipitation in preparation for protein assay. Neither trichloroacetic acid (TCA) nor phosphotungstic acid (PTA) precipitated latex glove proteins completely and a combination of both acids was required to achieve this. The precipitation procedure offers the opportunity - without substantial laboratory manipulation - to concentrate the protein in the leachate by redissolving the precipitate into a smaller volume of solvent thus increasing the sensitivity of protein detection. Each step of latex glove protein quantitation viz leaching precipitation concentration and assay has been shown to be quantitative. Applying TCA/PTA precipitation in conjunction with the micro-Lowry assay microgram and by extrapolation sub-microgram quantities of latex glove proteins can be detected and quantitated.
