Evaluation of plant growth, biomass, total phenolic content (TPC), and DPPH free radical scavenging activity of Marantodes pumilum at different ages of maturity and growing location

Abstract

Marantodes pumilum or locally known as kacip fatimah is an indigenous plant of Southeast Asia. Nowadays, it become one of the valuable medicinal herbs in Malaysia as it has been reported that phenolics from this herb contribute to a pharmacological activity that is effective for various illnesses. This study was conducted to evaluate the total phenolic content (TPC) and its antioxidant potential produced from three planting locations. For integrated planting, 30 plants of kacip fatimah were planted under rubber trees at Kg. Sagil, Ledang, Johor. The planting plots were accommodated with 70% shades using black netting and irrigated by the sprinkler system. For natural planting, about 30 plants of kacip fatimah were planted under the forest canopy at Forest Research Institute Malaysia (FRIM). The third planting was under the shade in FRIM’s Nursery located in Kepong Selangor. Evaluation for phenolic content was carried out at three, six, and nine months after planting (MAP). Leaves from ten plants of kacip fatimah were harvested at three months intervals and were soaked with 95% ethanol for 48 hours to proceed with crude and total phenolic content determination using a Follin-Ciocalture reagent. DPPH radical scavenging assay also was carried out to determine the antioxidant potential of planted kacip fatimah. Results show that the mean value for TPC and DPPH were inconsistently inconsistent at 3, 6, and 9 months after planting. The mean value of TPC from integrated planting is higher than the other two locations and the mean value of DPPH from natural planting is higher than the other two locations. Total phenolic content from integrated planting begins with 4841 ± 283.3 mg GAE /100g at 3 MAP, then reduced to 4219 ± 62.3 mg GAE /100g at 6 MAP and increases again at 9 MAP for 4504 ± 102.9 mg GAE /100g. The findings from this study will provide some patterns of secondary metabolite production that can be used by cultivators and the herbal industry to determine the ideal harvesting period and either to be planted at the nursery stage or by field planting.
