Abstract

Betel Piper betle L. is a medicinal plant with multiple traditional applications which originated from Peninsular Malaysia. It is locally known as sirih and reported to posses various beneficial bioactivities including anti-carcinogenic anti-inflammatory antioxidant and antimicrobial. The study was aimed to determine the optimum solid-liquid extraction SLE parameters including extraction temperature T and ratio of solvent to solid R for betel leaves. The optimization was carried out by considering the quality of extract and yield of SLE. The quality of extract was evaluated by comparing the contents of its active phytochemicals namely hydroxychavicol He and eugenol EU which were quantified with High Performance Liquid Chromatography HPLe. The effects of T and R on the yield of SLE were studied using response surface methodology RSM. The HPLC results indicated that the concentration of HC in the extracts increased with the extraction temperature. The percentage of increase dropped after 60C because the system was approaching the equilibrium in which saturation of HC happened. On the other hand the concentration of EU increased when the extraction temperature was raised from 25 to 60C but decreased as higher temperatures 70 and 80C were applied. A second-order polynomial model with regression coefficient R2 of 0.94 was solved using RSM for predicting of yield of SLE. The analysis showed that temperature exhibited greater effect on the yield compared to ratio of solvent to solid. The optimum temperature and ratio of solvent to solid were determined as 60C and 30:1 ml:g.
