Abstract
Bacteriophages of Ralstonia solanacearum were isolated from diseased hosts i.e. brinjal Solanum melongena chilli Capsicum annuum bell pepper Capsicum annuum var. grossum and Irish potatoes Solanum tuberosum; and the phages were designated as 22002 23040 23051 and 25007 respectively. Plaques of all phages were visible after incubation for 12 hours even though halo when present were only observed after 48 hours or 60 hours incubation. Sizes of plaques for each phage were not uniform in size. All phages varied in their adsorption rates with time. Phages had similar growth curve even though their latent period varied. Temperature studies showed that the optimum temperature for replication of phage 22002 was 30 DC while phage 25007 was 25C. Replication of phages 23051 and 23040 were not significantly different at both the temperatures. With the exception of phage 23040 all phages were reduced at 500 C for 10 minutes and were partially inactivated at 500 C for 60 minutes. However at 600 C all phages were almost completely inactivated when tested after 60 minutes. All phages were specific to strains of the pathogen. None could infect other genera or species of Bacillus spp. Pseudomonas fluorescens P. aeruginosa Erwinia carotovora E. chrysanthemi and Pseudomonas spp. commonly isolated from soils.
