Abstract

Heavy metals such as lead and zinc are abundant in the aquatic environment and affect the life of the organisms including free-living amoebae both at cellular and genomic level thus lead to a decline of their population size. This study examined the effect of zinc and lead on the DNA of Acanthamoeba polyphaga CCAP 1501/3A using comet and DNA laddering assays. The amoeba was exposed to a series of concentrations of zinc and lead in 24-well plates for 72 h before the amoeba viability and the IC‚‚ values of the metals against the amoeba were determined. The viability of amoebae was decreased in response to the increasing of heavy metals concentrations. The IC‚o value of zinc against this amoeba was 52.0 ppm and for lead was 15.0 ppm. Assessment done by comet and DNA laddering assays demonstrated that both heavy metals affect the amoeba both at its cellular and genomic level and induce apoptosis type of cell death.
