Abstract

Mussel DNA was successfully extracted using a CTAB-based method. Two Single Primer Amplification Reaction SPAR techniques namely Random Amplified polymorphic DNA RAPD and Random Amplified Microsatellites RAMs were used to study the population genetic structure of the green-lipped mussel. Perna viridis along the west coast of Peninsular Malaysia. One hundred and sixty five mussels were collected from Tanjung Rhu 20 Pulau Aman 10 Bagan Tiang 20 Muar 15 Anjung Batu 20 pontian 20 pantai Lido 20 Sg Belungkor 20 and Kg. Pasir Puteh 20. Total sixty eight markers were generated by RAPD 31 markers and RAMs 37 markers. Genetic distances were calculated based on Nei and Li;s 1979 coefficient. Three cluster analyses employing UPGMA were done based on RAPD markers RAMs markers and pooled RAPD and RAMs makers respectively. The cluster analysis based on RAPD markers grouped the populations into two major clusters. The Pulau Aman Tanjung Rhu Bagan Tiang pantai Lido Telok Emas Muar Anjung Batu and Kg Pasir Puteh populations were in one cluster while the Sg. Belungkor population clustered by itself. The RAMs cluster was different when compared with the RAPD cluster. The cluster analysis based on the RAMs markers showed that all the populations were clustered together while the cluster analysis based on pooled data from RAPD and RAMs markers showed a result similar to the RAPD based dendrogram. The dendrogram based on the RAPD markers appeared to be in accordance with the geographical distribution of the populations while the dendrogram based on the RAMs markers could be related to the reported pollution status of the sampling sites as noted in Table 2.
