Abstract
Mature pollen of oil palm is bi-nucleate with one vegetative and one generative cell. During pollen germination and pollen tube growth both nuclei move into the pollen tube where the generative nucleus divides to produce two sperm cells. Boron is an essential micronutrient involved in pollen tube growth. The effects of boron were studied in in vitro germination of pollen produced from oil palms of differing ploidy levels. Boron was applied as boric acid to a liquid pollen germination medium at concentrations ranging from O to 80 mM. Cytological examination of pollen germination and pollen tube growth were performed after 3 hours of adding mature pollen to the culture medium. Three different assessment methods were used: 1 vital staining FDA; 2 DNA staining using fluorescence detection from DAPI; and 3 aceto-orcein staining of nuclei. In the absence of borate pollen from diploid and triploid oil palm exhibited low germination rates which was associated with low fertility. This contrasted with a high rate in the presence of 1.6 mM boric acid. Borate concentrations failed to have an impact on pollen from haploid oil palm. The addition of borate to the germination medium was found to be beneficial and increased pollen fertility by up to 15 and provided optimal conditions for measuring pollen fertility in oil palm.
