Abstract
From the various organisms isolated three were found to be actively cellulolytic and they were identified as belonging to the genus Aspergillus denoted as Aspergillus 1 2 and 3 in descending order of their levels of cellulase activities. The optimum temperature for cellulase production of these Aspergillus species was found to be 37 deg C. Maximum enzyme production under the conditions studied was reached after an incubation period of 19 days for Aspergillus 1 and 3 and 15 days for Aspergillus 2. The cellulases of these isolates were most active at 37 deg C and within the pH range of 4.5 - 5.3. The enzymes from Aspergillus 2 and 3 were more stable than the enzyme from Aspergillus 1 at temperatures up to 45 deg C for 3 hours. Little or no activity remained at 60 deg C in all of the cases. The cellulases from these sources were stable in the pH range of 2.8 - 5.5. The enzyme from Aspergillus 2 had lowest Km value of 25 mg/ml Vmax 13.33 units/mg protein compared to the Km values of 40.0 mg/ml Vmax 8.33 units/mg protein and 28.6 mg/ml Vmax 11.11 units/mg protein for the cellulase of Aspergillus 1 and 3 respectively.
