Abstract

At least three fractions of cellobiohydrolases FPases EC 3.2.1.91 were partially purified from the culture filtrate of Aspergillus terreus SUK-1 through Ammonium Sulphate precipitation gel filtration chromatography Sephadex G150 and anion exchange chromatography DEAE-Sephadex A25. The three enzymes preparation namely as FPase I II and III were shown to be nearly homogenous on the 7.5 native polyacrylamide gel. There is no significant synergism observed between FPase and CMCase. However the activities of the combinations of FPase I - FPase II and FPase II - FPase III were synergistic to any substrate used in the assay. No definite ratio among the enzymes for maximal synergism could be established. The activities with FPase I-FPase III showed no synergism and the substrates used in order of high degree of synergism were carboxymethyl cellulose newspring filter paper and a-cellulose.
