Sperm concentration determination: 2X-Cel slide CASA vs. haemocytometer

Abstract

A total of 32 semen samples collected from 8 breeding bulls were used to compare Computer Assisted Semen Analysis CASA technique using a 2X-Cel slide with an Improved Neubauer haemocytometer gold standard for determination of sperm concentration. Collection of semen was made by electroejaculation method with/without rectal massage. Each semen sample was subjected to both techniques to determine the sperm concentration. Results showed a positive correlation r 0.54 between the two methods but with a significant difference P 0.05 in the measure of average sperm concentration for all samples. In the majority of the samples collected higher estimation of sperm concentration in millions per ml was observed in using CASA method than the conventional haemocytometer MSD 859536 vs. 525326. In conclusion 2X-Cel CASA showed a weak agreement with Haemocytometer for sperm count. CASA with a 2X-Cel slide may be used advantageously for a faster and objective determination of sperm concentration provided that further evaluation is made to determine the appropriate dilution range of raw semen to obtain a loser agreement with haemocytometer.
