Abstract
Three fern extracts from Blechnum orientale Diplazium esculentum and Stenochlaena palustris and the compound kaempferol-3-O-rutinoside were used to observe their effects on mitosis in root tip cells of Allium cepa and Commelina nudiflora. It was observed that at various dilutions the fern extracts and kaempferol-3-O-rutinoside caused cell division abnormalities in Allium cepa and Commelina nudiflora root tip cell. The abnormalities observed were such as bridges C-metaphase micronucleus disturbed anaphase and others. From a random total of 2800 cells observed in all dilutions from all extracts and kaempferol-3-O-rutinoside 20 per cent of the cells showed cellular division abnormalities. Apart from this silver staining of the nucleolar organising region NOR in the root tip cells during interphase were carried out to detect the level of activity of ribosomal genes. This was carried out by measuring the size and density of acidic non histone proteins transcribed by the genes in the NOR. Statistical analysis of variance was done to test the effects of the various dilutions of the diferent fern extracts and kaempferol-3-O-rutinoside on the mean silver stained NOR AgNOR size and the mean AgNOR density. The analyses showed kaempferol-3-O-rutinoside at various dilutions on the cellular division as well as the AgNOR size and density were highly significant.
