Abstract

Hevea brasiliensis latex beta-l 3-glucanase is an allergenic protein designated the allergen name Hevb 2 by the International Union of Immunological Societies and the World Health Organisation. The cDNA encoding the mature Hev b 2 protein of cultivar RRIM 600 was isolated by RT-PCR and cloned. The protein predicted from the DNA sequence has 316 amino acids. Its deduced molecular weight is 35 260 Da while its isoelectric point is estimated at 9.46. The recombinant fusion protein of Hev b 2 linked to the maltose-binding protein was synthesised in Escherichia coli. Recombinant Hev b 2 was recognised by the monoclonal antibody developed against the native protein but it was not reactive with IgE from subjects sensitised to Hev b 2. The implications of this observation in relation to the nature of IgE binding and to the development of latex immunoassays are discussed.
