Mapping of tissue culture related traits in oil palm (Elaeis guineensis Jacq.)

Abstract

The advent of DNA-based marker technology has enabled the location of marker-based quantitative trait loci (QTL) on genetic maps. The combined information of the molecular and phenotypic data of a mapping population facilitates the estimation of the putative location and effects of QTLs for traits of economic interest. Attempts were made in this study to map tissue culture amenability traits of oil palm including time to first callusing, average callusing rate, time to first embryogenesis and average embryogenesis rate as QTLs. A total of 87 F₁ palms of a dura x pisifera (progeny P2) mapping population was sampled for the purpose of tissue culture and map construction. The tissue culture data gave good segregation profiles for the traits of interest. Marker data were generated from 87 F₁ palms, and 2 parental maps were constructed, one for the dura (ENL48) and one for pisifera (ML161) parent. The parental map of dura has 143 markers and was resolved into 19 linkage groups and 5 marker-pairs. The total map distance for dura parent was 787cM with an average distance between 2 markers was 5.5cM. A total of 292 markers were resolved into 21 linkage groups with 3 marker-pairs on the pisifera map. The total genetic distance covered was 1416cM. The mean distance between markers was 4.8cM QTL analysis was carried out for both parents using interval mapping. QTLs were successfully estimated for the traits of interest in both the dura and pisifera parental maps.
